微环境中的代谢变化显著调节肿瘤免疫敏感性,但其潜在机制仍不清楚。最近,清华大学生命科学学院团队在期刊《Cell Metabolism》(IF=31.373)上发表题为“Cancer-cell-derived fumarate suppresses the anti-tumor capacity of CD8+ T cells in the tumor microenvironment.”的文章,报道了富马酸水合酶(FH)缺失的肿瘤表现出对功能性CD8+ T细胞活化、扩增和功效的抑制,并具有增强的恶性增殖能力。推断肿瘤细胞中的FH缺失会在肿瘤间质液中积累延胡索酸,增加的延胡索酸可直接在C96和C102处琥珀酸化ZAP70,并减弱其在浸润CD8+ T细胞中的活性,导致CD8+ T细胞活化受到抑制,并在体外和体内产生抗肿瘤免疫反应。此外,通过增加FH表达来消除延胡索酸盐可显著增强抗CD19 CAR T细胞的抗肿瘤功效。该研究证明了富马酸在控制TCR信号中的作用,并表明肿瘤微环境(TME)中的富马酸积累是CD8+ T细胞抗肿瘤功能的代谢障碍。该研提出了延胡索酸的减少可能是肿瘤免疫治疗的一个重要策略。
本文中使用了欣博盛Human Granzyme B ELISA kit检测了细胞培养上清中这个细胞因子的含量。
