近期，上海交通大学药学院沈琦团队在Advanced Functional Materials（IF=18.5）上发表题为“Self-Adjuvanting Bacteria Hydrogel for SHP1 Checkpoint Inhibition in Tumor-Draining Lymph Nodes to Enhance Cancer Immunotherapy.（自佐剂细菌水凝胶抑制肿瘤引流淋巴结SHP1检查点增强癌症免疫治疗）”的研究。尽管免疫疗法最近取得了进展，但其效果仍受到缺乏免疫协调的限制。特别是肿瘤引流淋巴结(TDLNs)与肿瘤之间的相互作用经常被忽视。在该项研究中，团队开发了一种能够激发强大且持久免疫反应的自佐剂水凝胶。该AAM水凝胶系统通过抑制肿瘤引流淋巴结中的SHP1免疫检查点，并增强肿瘤细胞因子效应，协同诱导抗肿瘤免疫反应，为安全而有效的抗肿瘤免疫治疗提供了一种创新策略。

表达IL-15的工程化的阿拉伯糖反应细菌(ARB)和负载维生素E的甘露糖修饰的中空介孔普鲁士蓝纳米颗粒(NP)(Man/HMPB(VE),MHV)与阿拉伯糖水凝胶(AraGel)混合，形成AraGel@ARB/MHV(AAM)系统。通过MHV介导的温和光热疗法，免疫原性细胞死亡(ICD)触发肿瘤相关抗原的释放。随后，Man修饰的NP靶向TDLN并释放VE，VE会抑制树突状细胞中含检查点Src同源区2结构域的磷酸酶-1(SHP1)，从而增强抗原呈递和T细胞的激活。同时，AraGel的降解会诱导ARB(IL-15)表达IL-15，这使得ARB能够以自佐剂的方式作为增强佐剂，与ICD和TDLN重编程协同作用，促进细胞毒性T淋巴细胞的激活。该水凝胶系统通过以协同的方式激发持久且强大的免疫治疗，有效地抑制了肿瘤的生长。总而言之，自佐剂水凝胶在癌症免疫治疗方面具有巨大潜力。
