近期,温州医科大学的沈建良课题组、蔡晓军课题组与邓辉课题组合作在《Advanced Science》(IF=15.1)上发表题为“Cascade Reactions Catalyzed by Gold Hybrid Nanoparticles Generate CO Gas Against Periodontitis in Diabetes.”(金混合纳米粒子催化级联反应产生一氧化碳气体对抗糖尿病牙周炎)的研究,为未来治疗糖尿病牙周炎提供了有益的启示。


由于糖尿病牙周炎存在局部炎症,其特点是葡萄糖浓度过高、细菌感染和高氧化应激,因此治疗糖尿病牙周炎是一项重大挑战。在该项研究中,研究团队将介孔二氧化硅纳米颗粒(MSN)缀合了金纳米颗粒(Au NPs)并负载了羰基锰,从而制备出一种一氧化碳(CO)增强型多酶协同杂化纳米平台(MSN-Au@CO)。Au NPs的类葡萄糖氧化酶活性可催化葡萄糖氧化成过氧化氢(H2O2)和葡萄糖酸,然后通过类过氧化物酶活性将 H2O2 转化为羟自由基(-OH),从而消灭细菌。此外,H2O2产生的CO与Au NPs对受到脂多糖攻击的巨噬细胞具有协同抗炎作用。其根本机制可能是诱导核因子红细胞2相关因子2(nuclear factor erythroid 2-related factor 2)减少活性氧,并抑制核因子kappa-B信号以减轻炎症反应。重要的是,MSN-Au@CO的抗菌和抗炎作用在患有结扎诱发的牙周炎的糖尿病大鼠模型中(ligature-induced periodontitis)得到了验证,证实了牙周骨质流失减少且具有良好的生物相容性。总之,MSN-Au@CO利用葡萄糖激活级联反应消灭细菌,并与气体疗法(gas therapy)协同调节免疫微环境,为糖尿病牙周炎的治疗提供了一个潜在的方向。
