经典的(caspase-1)和非经典的(包括caspase-4、5和11)在炎症小体中均切割gasdermin D (GSDMD)诱导细胞焦亡。caspase-1加工处理IL-1β和IL-18使其成熟 , 而目前还没有确定脂多糖激活的caspase-4、5和11细胞因子靶点。最近,北京生命科学研究所研究团队在期刊《Nature》(IF=64.8)上发表题为“Recognition and maturation of IL-18 by caspase-4 noncanonical inflammasome.”的文章,该研究团队的研究结果表明显示激活的人caspase-4,而不是小鼠 caspase-11在体外和细菌感染过程中直接加工IL-18 。caspase-4切割pro-IL-18中与caspase-1相同的四肽位点。 caspase-4-pro-IL-18复合物的晶体结构揭示了一个双位点(二元) 底物识别机制。这种二元识别也被caspase-5和caspase-1用来加工pro-IL-18。这项研究发现并证明炎症因子IL-18是非经典炎症小体通路活化的caspase-4/5的底物,并完整地揭示了天然免疫通路中caspases识别和切割pro-IL-18,以及pro-IL-18被加工后成熟为有生理功能的炎症因子的精确分子机制,为理解一些自身免疫疾病的发生机制以及开发新的治疗方法提供了全新的思路。
本文中使用了欣博盛Human IL-18和 IFN-γ ELISA kits 检测了细胞培养液上清中目标蛋白的含量。
