动脉粥样硬化是许多心血管疾病中常见的病理现象。尽管目前的疗法(包括他汀类药物和丝氨酸蛋白酶PCSK9抑制剂)可以有效地将低密度脂蛋白(LDL)胆固醇水平降低到临床指南推荐的水平,但心血管不良事件仍然频繁发生。脂蛋白在动脉壁的内皮下滞留触发了巨噬细胞的多种炎症事件,是动脉粥样硬化病理进展的主要原因。因此,调节炎症是预防和治疗动脉粥样硬化及其并发症的一种途径。不幸的是,在控制斑块炎症的特异性和有效性方面的挑战阻碍了动脉粥样硬化治疗的进展。最近,上海交通大学研究团队在期刊《ACS nano》(IF=17.1)上发表题为“Modulating Plaque Inflammation via Targeted mRNA Nanoparticles for the Treatment of Atherosclerosis”的文章,报道了一种NP介导的mRNA治疗方法,以动脉粥样硬化病变巨噬细胞为靶点,调节晚期动脉粥样硬化病变中的炎症,从而治疗动脉粥样硬化。证明了含有IL-10 mRNA的靶向NPs与M2样巨噬细胞共定位,并在静脉注射给Western diet(WD)喂养的Ldlr小鼠后诱导动脉粥样硬化斑块中IL-10的产生。此外,氧化应激和巨噬细胞凋亡减少证明损伤显示出炎症反应显著减轻,导致脂质沉积减少,坏死面积减少,纤维帽厚度增加。这些结果表明,在晚期动脉粥样硬化的临床前模型中,将mRNA治疗剂成功递送至富含巨噬细胞的斑块,表明这种靶向NP炎症方法具有转化为广泛临床应用的巨大潜力。
本文中使用了欣博盛Mouse IL-10, IL-1β和 IL-6 ELISA kits检测了主动脉组织和血清样本中目标蛋白的含量。
